Summary

Ziele im hier vorgestellten Projekt sind die Entwicklung hocheffizienter und kostengünstiger Nachweismethoden zur sicheren und schnellen Bestimmung des Befalls mit Tilletia Arten sowie Ustilago nuda bei Weizen und Gerste. Diese Methoden sollen neben der Bestimmung der Befallshöhe eine eindeutige Identifizierung der Pathogene, insbesondere auch bei Mischinfektionen von T. caries und T. controversa, ermöglichen. Dieses Ziel soll durch die Entwicklung und Optimierung von q-PCR und LAMP basierten Methoden erreicht werden. Die neu entwickelten Methoden werden in umfangreichen Validierungsstudien hinsichtlich Genauigkeit, Robustheit, Reproduzierbarkeit und Wiederholbarkeit überprüft und verglichen. Anschließend soll die Aufnahme in das Methodenbuch der ISTA erfolgen. Um die gesetzten Ziele zu erreichen, werden im ersten Jahr die q-PCR an der LfL und die LAMP-Technologie am JKI für T. caries, T. controversa, T. indica und Ustilago nuda etabliert. Da für die q-PCR als auch für die LAMP-Technologie z. T. die gleichen Primer verwendet werden können, erfolgt auch eine gemeinsame Primerentwicklung und Validierung, um alle Synergien in der Methodenentwicklung auszuschöpfen. Im zweiten Jahr finden v. a. Validierungsstudien bezüglich der Robustheit des Nachweises der unterschiedlichen Tilletia-Arten und Ustilago nuda sowie der Quantifizierung statt. Es werden verschiedene DNA-Isolationsmethoden getestet und mögliche Kreuzreaktionen mit anderen Getreidepilzen untersucht. Im dritten Jahr werden an der LfL neben weiteren Validierungsstudien, die Umsetzung größerer Multiplex-Ansätze, und der Vergleich der entwickelten Methoden forciert. Die Methoden mit der größten Robustheit, Reproduzierbarkeit und Wiederholbarkeit werden zur Aufnahme in das ISTA-Methodenbuch vorgeschlagen. Parallel dazu erfolgt am JKI die Untersuchung der Populationsstruktur von T. caries und T. controversa anhand bereits publizierter und neu zu entwickelnder molekularer Marker (Mikrosatelliten).


Summary translation

In the here presented project highly efficient and economically viable methods will be developed to detect seed born smut fungi (Tilletia spp., Ustilago nuda) in wheat and barley. With the help of these methods we are not only aiming at individually and unambiguously identifying the pathogens, also in mixed contaminations of seed lots, but we are also aiming at quantifying the contamination level of seed lots by these fungi. To reach these goals molecular biological methods, based on q-PCR and LAMP, respectively, will be developed and optimised. These methods will be internationally validated and then submitted to the International Seed Testing Association (ISTA) for implementation in the handbook 'International Rules for Seed Testing'. In the first year q-PCR methods will be developed and optimised at the Landesanstalt fu¨r Landwirtschaft (LfL) while LAMP will be established at the Julius Ku¨hn-Institut (JKI). As far as possible primer development and validation of the methods will be performed jointly to take advantage of potential synergies. In the second year validation studies in both institutions LfL and JKI will focus especially on the accuracy and robustness of identification of the different smut fungi and on their quantification. Several DNA-isolation-methods will be tested and potential cross-reactions of the developed methods with other fungal cereal parasites will be investigated. In the third year, more validation studies covering especially reproducibility, and repeatability will be performed at the LfL. Multiplexing of the tests will also play an important role. At JKI population structure of selected T. caries and T. controversa populations will be studied using already published and newly developed molecular markers (microsatellites) for these fungi.
