Summary

Rhizoctonia solani Kühn ist ein weit verbreiteter bodenbürtiger Erreger, der an verschiedenen Kulturen für ökonomisch relevante Ertragsverluste verantwortlich ist. Der Erreger R. solani ist aufgrund seiner sowohl saprophytischen als auch parasitischen Eigenschaften sowie seiner persistenten Dauerorgane (Sklerotien) schwer zu bekämpfen. Seine Fähigkeit auch an Nichtwirtspflanzen seinen Lebenszyklus vollenden zu können, erschwert die Kontrolle über Fruchtfolgen, die insbesondere im organischen Landbau Teil der Bekämpfungsstrategie gegen bodenbürtige Erreger sind. Probleme durch R. solani treten im ökologischen Anbau vor allem an Kartoffeln auf. Derzeit stehen dem ökologischen Anbau keine geeigneten Strategien zur Bekämpfung des Erregers zur Verfügung. Ziel des Projektes ist daher die Entwicklung einer biologischen Methode zur Kontrolle des Erregers auf der Basis eines bakteriellen und pilzlichen Antagonisten. Durch Einsatz eines bakteriellen Antagonisten soll die Widerstandsfähigkeit der Pflanze gegenüber R. solani erhöht und durch Anwendung eines pilzlichen Antagonisten die Dauerorgane im Boden oder an Kartoffelknollen bekämpft werden.
Im Rahmen des Projektes wurden 450 bakterielle und 390 pilzliche Isolate auf der Basis eines hierarchischen Screeningsystems aus verschiedenen in vitro und ad planta Methoden hinsichtlich ihrer Eignung als Biological Control Agents (BCAs) zur Kontrolle von R. solani geprüft. Insgesamt konnten durch in vitro Untersuchungen 18 bakterielle und 12 pilzliche Isolate gegen das Zielpathogen selektiert werden. Das Myzel von R. solani wird von den pilzlichen Antagonisten parasitiert und die Sklerotienkeimung vollständig gehemmt. Ihre Wirkung wurde bislang an natürlich mit Rhizoctonia-Sklerotien infizierten Kartoffelknollen, künstlich infizierten Kartoffelknollen sowie Salatpflanzen und Zuckerrübensämlingen unter krankheitsbegünstigenden Bedingungen geprüft. In diesen Versuchen zeigten insbesondere 3 bakterielle (Pseudomonas putida B1, Pseudomonas fluorescens B2 und Serratia plymuthica B4) und 6 pilzliche (P2, P3, P4, P9, P10, P11, Trichoderma spp.) Antagonisten eine wiederholte, signifikante krankheitsunterdrückende Wirkung, z. B. bis zu 60 % durch B1 oder über 50 % durch P11 an Kartoffelkeimlingen.
Unter Feldbedingungen wurde der marktfähige Kartoffelertrag durch R. solani bei einem hohen Befalldruck signifikant reduziert und der Befall der Knollen mit R. solani Sklerotien signifikant erhöht. Durch die Bakterisierung der Pflanzkartoffeln konnte die Befallsschwere signifikant reduziert und der Ertrag erhöht werden. An Salat war ebenfalls eine Kompensation der durch R. solani verursachten Biomasseverluste gegeben. Die Ergebnisse erster Feldversuche mit den bakteriellen Antagonisten B1, B2 und B 4 an Salat und Kartoffel bestätigten somit deren krankheitsunterdrückende Wirkung.
Durch eine optimale Applikationsdichte und –häufigkeit sowie die Entwicklung einer geeigneten Formulierung für die gegen R. solani wirksamsten BCAs (bakteriell sowie pilzlich) könnte die krankheitsunterdrückende Wirkung verbessert und eine höhere Wirksicherheit gewährleistet werden. Dies ist in weiteren Feldversuchen zu prüfen.


Summary translation

The soil-borne pathogen Rhizoctonia solani Kühn is worldwide responsible for serious damage of many economically important agricultural and horticultural crops. Control of Rhizoctonia diseases is difficult because this pathogen survives for many years as sclerotia in soil or as mycelium in organic matter under numerous environmental conditions. Furthermore, the pathogen has an extremely wide host range and is able to colonise plant species, even those not recognised as usual R. solani host plants, and thereby to complete its life cycle. In fact, lack of knowledge of ‘non-host plants’ reduces the potential of crop rotation, which is an important control strategy against soil-borne pathogens in organic farming. Problems by R. solani in organic farming exist especially on potato. To date, no effective control strategies against this pathogen are available. Hence, our work is concentrated on the development of a biological control method based on bacterial and fungal control agents. The bacterial antagonist should improve plant health, whereas the fungal antagonist control the pathogen in soil or the sclerotia on potato tubers.
In the project the antifungal activity of 450 bacterial strains and 390 fungal strains were evaluated toward R. solani based on a hierarchical combination of in vitro and in vivo assays. As result of different screening steps in vitro (antagonistic potential, cell wall-degrading enzymes), 18 rhizo-bacterial isolates and 12 fungal isolates were selected. Hyperparasitism of the R. solani mycel and an inhibition of sclerotia germination was to observed by the fungal antagonists. Bacterial and fungal strains with antifungal activity were further evaluated in vivo assays on naturally with Rhizoctonia infected potato tubers, artificial infected potato tubers, lettuce plants and sugar beet seedlings under favorable conditions for the disease development. As result of these experiments three potato-associated ecto- and endophytically living bacterial strains (Pseudomonas fluores-cens B1, P. fluorescens B2 and Serratia plymuthica B4) and six fungal strains (P2, P3, P4, P9, P10, P11, Trichoderma spp.) showed repeated disease suppression effects against R. solani on potato, lettuce and sugar beet. The strain B1 limited the black scurf symptoms on potato sprouts to 60 % and the fungal strain P11 to 50 %.
Under field conditions the marketable tuber yield on potato was reduced at high disease pressure by R. solani. Additionally, the proportion of sclerotia infested tubers increased statistically significantly at high disease pressure. Using bacterisation of seed tubers the disease severity caused by R. solani could be significantly reduced, and the marketable tuber yield could be increased. The biomass losses on field grown lettuce caused by R. solani could be compensated by bacterisation of lettuce seedlings. The results of field experiments with the bacterial antagonists B1, B2 and B4 supported the disease suppression effect of climate chamber experiments.
The development of a suitable formulation can improve the disease suppression effect of the bacterial and fungal antagonists. Furthermore, the effect of biocontrol strains will be tested in different field experiments in dependence on their application frequency and density.
