Summary

Zur Entwicklung ölreicher, krankheitstoleranter Saflor-Stämme wurde ein in der ehemaligen Thüringer Zuchtstation Bendeleben in den 1930er Jahren entwickelter Stamm (nur ca. 25 % Ölgehalt, aber gute Krankheitsresistenz) im Gewächshaus mit der spanischen Sorte ‚Sepasa’ (bis 46 % Ölgehalt, aber hohe Botrytisanfälligkeit) frei abblühen gelassen. Die Auskreuzung betrug ca. 50 %. Aus der im Feld als Einzelpflanzen angebauten F2-Generation wurden 550 ertragreiche Pflanzen geerntet und von jeder Pflanze an 30 Körnern der durchschnittliche Schalenanteil ermittelt. Pflanzen mit einem Schalenanteil < 40 % sind im Feld nachgebaut worden (ca. 50 Nummern, Einzelpflanzenanbau, 30 x 30 cm, 5,4 m² Anbaufläche/Nachkommenschaft). Je eine Pflanze mit hohem Kornertrag und niedrigem durchschnittlichen Schalenanteil aus jeder Parzelle wurde für den nächstjährigen Anbau ausgewählt und gleichzeitig das Restsaatgut der 14 besten Parzellen (homogen, frühreif, ertragreich) für eine exakte Ertragsprüfung verwendet.
Die Ermittlung des Schalenanteils an Einzelkörnern und –pflanzen hat sich als schnelle und preiswerte Methode zur Einschätzung des Ölgehaltes erwiesen (ermittelte Korrelation zwischen Ölgehalt und Schalenanteil r = 0,88). Technisch ist die Trennung von Schale und Kern leicht durch kurzzeitiges Einquellen der Samen in Wasser zu erreichen.
In den bisher fünfjährigen Ertragsprüfungen mit Stämmen aus der Kreuzung ‚Sepasa’ und Stamm ‚Bendeleben’ hat sich gezeigt, dass diese der Standardsorte ‚Sabina’ in günstigen Jahren mit sonnigem und trockenem Wetter während der Blüte ertraglich überlegen sein können. Sie wiesen gleichzeitig einen im Durchschnitt 30 % höheren Ölgehalt als ‚Sabina’ auf. Die Stämme waren in morphologischer Hinsicht bereits in der F4 weitgehend homogen, enthielten aber noch viele botrytrisanfällige Pflanzen, wie sich 2007 zeigte. In diesem Jahr mit starkem Infektionsdruck waren bei zwei Drittel aller angebauten Stämme und Einzelpflanzennachkommenschaften (EPN) gravierende Ertragseinbußen festzustellen. Immerhin wiesen 15 EPN aber durchgängig keine Ertragsminderung bei gleichzeitig höherem Ölgehalt als ‚Sabina’ auf. Es ist also durchaus möglich, durch Einkreuzung von südeuropäischen oder amerikanischen Stämmen, die sich durchgehend in eigenen Prüfungen als botrytisanfällig erwiesen haben, in angepasste Typen, botrytistolerante Formen mit hohen Ölgehalten zu entwickeln.
Als einfachste Methode zur Selektion von dünnschaligen, ölreichen Typen aus spaltenden Populationen hat sich die Isolation von ca. 20 Blütenköpfchen einer Pflanze mit Crispac-Tüten mit Supermicrolochung und Bestimmung deren genetischen Status an Einzelkörnern aus nichtisolierten Köpfchen derselben Pflanze erwiesen. Der Samenansatz unter Isolation ist allerdings gering und die Keimfähigkeit der Samen niedrig. Andere Methoden, wie die Auslese von Einzelpflanzen mit niedrigem Schalenanteil oder die Anzucht von schalenlosen Kernen mit hohem Ölgehalt führten nur zu einem partiellen Erfolg, so dass in den Ertragsprüfungen nur 37,6 % Ölgehalt bei einem Stamm erreicht werden konnten.
Die Probleme des Safloranbaus unter mitteleuropäischen Bedingungen sind mit der Entwicklung botrytistoleranter ölreicher Stämme nicht gelöst. Eine ausreichende Befruchtung ist in kühlen und feuchten Jahren nicht gegeben, weil Saflor zwingend auf die Bestäubung durch Insekten angewiesen ist. Es kommt zur Bildung einer größeren Anzahl parthenokarper (= hohler) Samen, wodurch der Ertrag und insbesondere der Ölgehalt deutlich sinken. Die Erhöhung der Selbstfertilität muss deshalb zukünftig ein wichtiges Zuchtziel sein.

To develop types of safflower with a high oil content and disease tolerance a strain of the former Thuringian breeding station Bendeleben with a good disease tolerance but an oil content about 25 % was crossed with the Spanish variety ‘Sepasa’ (up to 46 % oil content and high susceptibility to Botrytis) in a greenhouse by jointly free blossom. The outcrossing was about 50 %. 550 productive plants from the F2, cultivated as single plants in the field were harvested. The average hull content of each plant was determined on examination of 30 seeds. Plants with a percentage of hulls < 40 % were reproduced in the field in the next year (approx. 50 specimens) on 5.4 m² cultivation area per descendant, 30 x 30 cm plant distance. Out of each plot one plant with high seed yield and low percentage of hulls was chosen for the cultivation in the following year. The remaining seed of the 14 best plots (morphologic homogeneous, early ripening, high seed yield) was used for an exact field trial.
The determination of the percentage of hulls of single seeds or single plants has proved to be a quick and cheap method to estimate the oil content of safflower (determined correlation coefficient between oil content and percentage of hulls r = 0.88). The hulls easily can be separated from the nuclei by soaking the seeds in water for an short time.
The up to now within five years carried out yield trials with the cross-breeded strains of Stamm ‘Bendeleben’ and ‘Sepasa’ have shown that those can exceed the standard variety ‘Sabina’ in favourable years (sunny and dry weather during the flowering time) regarding the seed yield. Besides their oil content on average was 30 % higher than the oil content of ‘Sabina’. Already in the F4 they were nearly homogeneous considering the morphological aspects but still consisted many Botrytis-susceptible plants. This could be seen in 2007, a year with a high pressure of Botrytis infection. Two thirds of the cultivated strains (examination plots and single plant descendants) shown heavy reduction caused by Botrytis-infection. But still 15 single plant descendants have had similar yield per plant as in the year before without infection. Their oil content was higher as those of ‘Sabina’. So it is possible to develop types of safflower with a high oil content and tolerance against Botrytis by cross-breeding of local tolerance accessions with susceptible South-European or American varieties.
The most simple method for select oil-rich types from segregated populations seems to be the isolation of about 20 flower-heads of a plant with a Crispac-bag and the determination of their genetic status by examining about 20 single plants from non-isolated flower-heads of the same plant in regard to their percentage of hulls. Other methods like the selection of single plants with a low percentage of hulls or the cultivation of plants from hull-less, oil-rich nuclei only had a partial success so that only one strain showed an oil content of 37.6 % in the yield trials.
The problem with the cultivation of safflower under Middle-European conditions are not solved by the development of Botrytis-tolerant strains. Because the plant needs the help of insects for its fertilization there is not guaranteed a full pollination in wet, cool years. The consequence of this are low seed yield with a low oil content. Therefore, an aim of further investigations must be to increase the self fertility of safflower.
